All substrates were designed using HyCoSuL technology (Hybrid Combinatorial Substrate Library, Poreba et al., Nature Protocols, 2017, (10): 2189-2214) in which natural and unnatural amino acids are used to optimize protease specificity of the tetrapeptide substrates. All substrates are linked to an ACC fluorophore due to its 2.8 -fold higher fluorescence quantum yield at the excitation and emission wavelengths of 380 nm and 460 nm. This results in higher sensitivity of the assay.
